Anatomical and biochemical changes associated with the development and germination of Araucaria angustifolia seeds Biological Sciences
Shibata, Marília; Coelho, Cileide Maria Medeiros; Schmidt, Eder Carlos; Bouzon, Zenilda Laurita; Campos, José Marcello Salabert de; Maraschin, Marcelo.
This study aimed at assessing the cell cycle, and anatomical and biochemical changes that the embryonic axis of Araucaria angustifolia undergoes during development, focusing on the maturation stage. During all development, cells exhibited intense metabolic activity with an abundance of mitochondria, lipid bodies, and vacuolated cells. The continued accumulation of starch and protein was observed by LM and TEM and indicated by spectra of FTIR. Cell differentiation of the procambium was observed with a thickening of the cell wall and the formation of resiniferous ducts. At Stage III and IV, cells exhibited structural changes such as altered or elongated mitochondria and presence of plastoglobules. These results suggest that there is a gradual transition from...

Harvest time and agronomic performance of maize hybrids with contrasting growth cycles Agronomy
Panison, Fernando; Sangoi, Luís; Kolling, Daniel Fernando; Coelho, Cileide Maria Medeiros; Durli, Murilo Miguel.
The delay of the maize harvest may cause qualitative and quantitative grain losses. This work evaluated the effects of the harvest time on the agronomic performance of maize hybrids with contrasting growth cycles. The experiment was set in Lages, Santa Catarina State,South Brazil, during the 2012/2013 and 2013/2014 growing seasons. A randomized block design, arranged in split plots was used. Five single-cross hybrids were tested in the main plots: P1630H and P32R22H (hyper-early), P2530 (super early) and P30F53YH and P30R50YH (early). Five harvest times were assessed in the split plots: 0 (grain physiological maturity), 10, 20, 30 and 40 days after physiological maturity. The sowing was performed on 12/5/2012 and 10/5/2013. The percentage of lodged and...

Osmolyte accumulation and antioxidant metabolism during germination of vigorous maize seeds subjected to water deficit Agronomy
Prazeres, Camila Segalla; Coelho, Cileide Maria Medeiros.
The objective of this work was to evaluate the alterations of antioxidant enzyme reserves and antioxidant enzymes during germination under water deficit in maize hybrids and to associate with seed vigor, determining the mechanisms related to tolerance for this stress. Two three-way maize hybrids were characterized by their vigor at different levels of water deficit induced by polyethylene glycol 6000. Next, the seeds were hydrated at different osmotic potentials (0.0, -0.3, and -0.9 MPa) and removed at different times to assess the levels of the total soluble protein, total soluble sugars, proline, starch, and antioxidant enzymes, such as superoxide dismutase, catalase and ascorbate peroxidase. The analysis of variance, Tukey test at 5% and principal...

Physiological and physical quality of local Araucaria angustifolia seed variety Agronomy
Shibata, Marilia; Coelho, Cileide Maria Medeiros; Araldi, Cristhyane Garcia; Adan, Natalia; Peroni, Nivaldo.
Araucaria angustifolia seeds are the basis for the rural economy in southern Brazil, and the species presents different botanical varieties. The objective of this work was to evaluate the diversity of physical, physiological and biochemical characteristics of Araucaria angustifolia seeds from local varieties: ‘sancti josephi’, ‘angustifolia’, ‘caiova’, and ‘indehiscens’. Seeds were collected from the cities of Painel and Urubici in Santa Catarina State and tested for moisture content, 1000-seed weight, emergence, emergence speed index (ESI), root and shoot length, electrical conductivity, tetrazolium, soluble carbohydrates, and starch content. Among the varieties cited above, the angustifolia variety from Painel displayed a greater 1000-seed weight, ESI,...

Reserve metabolism of stored and germinated Araucaria angustifolia seeds Agronomy
Araldi, Cristhyane Garcia; Coelho, Cileide Maria Medeiros.
Germination metabolism of recalcitrant seeds of Araucaria angustifolia is activated in storage, which complicates the seeds conservation and utilization. This study aimed to identify the changes in the reserve metabolites of A. angustifolia seeds throughout storage in order to understand the processes of hydrolysis caused by germination metabolism. Mature seeds were harvested in southern Brazil and stored in an ambient environment and cold chamber conditions. Biochemical analyses were performed for embryos and megagametophytes from seeds stored for 0, 15, 30, 45, and 90 days. Due to seeds being in advanced germination in storage, they were evaluated at 90 days in different early developmental categories: I – seeds with mature embryos, II – seeds with...
